Details

Palonosetron blocks the actions of chemicals in the body that can trigger nausea and vomiting. Palonosetron is used in adults to prevent nausea and vomiting caused by surgery or by receiving medicine to treat cancer (chemotherapy). Palonosetron is used in children only to prevent nausea and vomiting caused by cancer medicine. Palonosetron is not for preventing nausea or vomiting that is caused by factors other than chemotherapy or surgery. Palonosetron is usually given 30 minutes before chemotherapy or right before you are given anaesthesia for surgery.


Typical Uses

Palonosetron is used to prevent nausea and vomiting caused by cancer drug treatment (chemotherapy). It is also used to prevent nausea and vomiting after surgery. Palonosetron works by blocking one of the body's natural substances (serotonin) that causes vomiting. This medication may also be used for the treatment of nausea and vomiting caused by chemotherapy.


Side Effects

Common palonosetron side effects may include high levels of serotonin in the body-agitation, hallucinations, fever, fast heart rate, overactive reflexes, nausea, vomiting, diarrhoea, loss of coordination, fainting.


Drug Interactions

  • Palonosetron+alfentanil- Using palonosetron together with alfentanil can increase the risk of a rare but serious condition called the serotonin syndrome, which may include symptoms such as confusion, hallucination, seizure, extreme changes in blood pressure, increased heart rate, fever, excessive sweating, shivering or shaking, blurred vision, muscle spasm or stiffness, tremor, incoordination, stomach cramp, nausea, vomiting, and diarrhoea.
  • Palonosetron+amoxapine- Using palonosetron together with amoxapine can increase the risk of a rare but serious condition called the serotonin syndrome, which may include symptoms such as confusion, hallucination, seizure, extreme changes in blood pressure, increased heart rate, fever, excessive sweating, shivering or shaking, blurred vision, muscle spasm or stiffness, tremor, incoordination, stomach cramp, nausea, vomiting, and diarrhoea.
  • Palonosetron+bepridil- Using bepridil together with palonosetron can increase the risk of an irregular heart rhythm that may be serious and potentially life-threatening, although it is a relatively rare side effect. You may be more susceptible if you have a heart condition called congenital long QT syndrome, other cardiac diseases, conduction abnormalities, or electrolyte disturbances.
  • Palonosetron+desipramine- Using palonosetron together with desipramine can increase the risk of a rare but serious condition called the serotonin syndrome, which may include symptoms such as confusion, hallucination, seizure, extreme changes in blood pressure, increased heart rate, fever, excessive sweating, shivering or shaking, blurred vision, muscle spasm or stiffness, tremor, incoordination, stomach cr


Mechanism of action

Palonosetron belongs to a class of antiemetic medications called serotonin (5HT3) antagonists. It binds to special sites called serotonin 5HT3 receptors and blocks the action of serotonin, a naturally released chemical in the body that triggers nausea and vomiting.
