Insulin is a hormone that is produced in the body. It works by lowering levels of glucose (sugar) in the blood. Insulin aspart and insulin aspart protamine is a faster-acting form of insulin than regular human insulin. Insulin aspart and insulin aspart protamine is a combination medicine used to treat type 1 (insulin-dependent) diabetes in adults. To make sure this medicine is safe for you, tell your doctor if you have liver or kidney disease. You should not use this medicine if you are allergic to insulin, or if you are having an episode of hypoglycaemia (low blood sugar).


Typical Uses

Insulin aspart is used with a proper diet and exercise program to control high blood sugar in people with diabetes. Controlling high blood sugar helps prevent kidney damage, blindness, nerve problems, loss of limbs, and sexual function problems. Proper control of diabetes may also lessen your risk of a heart attack or stroke. Insulin aspart is a man-made product that is similar to human insulin. It replaces the insulin that your body would normally make. Insulin aspart starts working faster and lasts for a shorter time than regular insulin. It works by helping blood sugar (glucose) get into cells so your body can use it for energy. This medication is usually used with a medium- or long-acting insulin product.


Side Effects

Common side effects of insulin aspart include vision changes swelling in your hands or feet itching, swelling, redness, or thickening of the skin where you injected the medicine or low potassium-confusion, uneven heart rate, extreme thirst, increased urination, leg discomfort, muscle weakness or limp feeling.


Drug Interactions

  • Insulin aspart+gatifloxacin- Gatifloxacin can affect blood glucose levels and should not be used in patients with diabetes. Both hypoglycaemia (low blood glucose) and, less frequently, hyperglycaemia (high blood glucose) have been reported. Severe cases of hypoglycaemia during treatment with gatifloxacin have resulted in coma and even death. You should learn to recognize the symptoms of hyperglycaemia and hypoglycaemia, as well as what to do when these conditions occur.


Mechanism of action

Insulin aspart protamine binds to the insulin receptor (IR), a heterotetrameric protein consisting of two extracellular alpha units and two transmembrane beta units. The binding of insulin to the alpha subunit of IR stimulates the tyrosine kinase activity intrinsic to the beta subunit of the receptor. The bound receptor autophosphorylates and phosphorylates numerous intracellular substrates such as insulin receptor substrates (IRS) proteins, Cbl, APS, Shc and Gab 1. Activation of these proteins leads to the activation of downstream signalling molecules including PI3 kinase and Akt. Akt regulates the activity of glucose transporter 4 (GLUT4) and protein kinase C (PKC).
